Inactivation of ribosomes in vitro by colicin E 3 .
نویسنده
چکیده
Cell-free protein synthesizing extracts incubated with purified preparations of colicin E(3) showed a marked decrease of their protein synthesizing activity. Incubation of ribosomes prepared from S-30 extracts with E(3) resulted in their inactivation. Their 16S RNA was found to lose a terminal fragment, similar in size to the fragment released upon in vivo treatment with E(3). Extracts derived from colicinogenic strains are resistant to the action of E(3)in vitro.

Stimulation of in vitro colicin E3 action by messenger ribonucleic acid and message analogs.
Ribosomes that are synthesizing polypeptides in vitro on certain natural and synthetic messages are more susceptible to inactivation by colicin E3 than ribosomes treated with E3 in the absence of added message. The initiation step of protein synthesis appears to be sufficient to cause the conversion of ribosomes to a state more sensitive to colicin E3. Message and protein synthesis are not, how...
متن کاملSpecific inactivation of 16S ribosomal RNA induced by colicin E3 in vivo.
Treatment of sensitive Escherichia coli cells with colicin E3 leads to inactivation of 30S ribosomal subunits. In vitro reconstitution of 30S subunits indicates that the E3-induced defect lies solely in the 16S RNA. 16S RNA from E3-treated cells lacks several T(1) RNase oligonucleotides of normal 16S RNA, including the one from the 3'-end of the 16S RNA, as analyzed by the fingerprint technique...
متن کاملDetection of Colicin genes by PCR in Escherichia coli isolated from cattle in Shiraz-Iran
A variety of probiotic bacteria have been tested to control animal and foodborne pathogenic bacteria in livestock. The mechanism of inhibition of pathogenic bacteria for several of those probiotic microorganisms is mediated by the production of bacteriocins. Colicins are probably the group of bacteriocins that have been most thoroughly characterized. Colicins are antimicrobial proteins produced...
متن کاملTranscriptional profiling of colicin-induced cell death of Escherichia coli MG1655 identifies potential mechanisms by which bacteriocins promote bacterial diversity.
We report the transcriptional response of Escherichia coli MG1655 to damage induced by colicins E3 and E9, bacteriocins that kill cells through inactivation of the ribosome and degradation of chromosomal DNA, respectively. Colicin E9 strongly induced the LexA-regulated SOS response, while colicin E3 elicited a broad response that included the induction of cold shock genes, symptomatic of transl...
متن کاملColicin E3 cleavage of 16S rRNA impairs decoding and accelerates tRNA translocation on Escherichia coli ribosomes
The cytotoxin colicin E3 targets the 30S subunit of bacterial ribosomes and specifically cleaves 16S rRNA at the decoding centre, thereby inhibiting translation. Although the cleavage site is well known, it is not clear which step of translation is inhibited. We studied the effects of colicin E3 cleavage on ribosome functions by analysing individual steps of protein synthesis. We find that the ...
